The Abnormality of Monoamine and 8-OHdG Concentration and Change of Anti-oxidant Enzyme Activities in the Brain of Mouse and Hamster Infected with Scrapie Agent

Abstract
Scrapie is a degenerative disease of central nervous system of sheep and a few other animals. Amyloid plaque and vacuolation which of the signs of senile dementia are found in the brain of scrapie-infected animals. The significant decrease of
aminergic
neurotransmitter may be a major cause of clinical symptom such as ataxia and head bubbing. So we tried to meassure the concentration of neurotransmitter with HPLC. The overall decrease of the concentration of adrenergic and serotonergic
neurotransmitter
was measured. The significant decrease of norepinephrine in cerebral cortex of 87-infected IM mouse brain and dopamine in brain stem of 263K-infected hamster was shown. The involvement of oxygen free radical in various degenerative disease was
thought
to play important role in processing of neurodegenerative disease, however, 8-OHdG, the indicator of DNA oxidation by free radicals, was not altered in number in the brain of serapie-infected animals. And the activities of antioxidant enzymes
located in
cytosol were not altered either, except that of glutathione reductase of hamster brain. Glutathione reductase is concerned with lipid peroxidation. So these results provide the possibility of relationship between neuropathological changes and
membrane
lipid peroxidation in scrapie agentinfected animals.
